Abstract:

This study was designed to evaluate the toxic effect of ciprofloxacin on the morphology of adult rat kidneys by light and electron microscope.It was conducted on twenty adult rats,they were divided into 2 groups,5 rats as a control group given pure distilled water and 15 rats as experimental group given oral ciprofloxacin.Each rat in experimental group received 500mg/kgm/body weight orally by gastric tube(single daily dose)which equal 1/10 LDS50 of the tested drug for four weekd.After this period ,rats of both groups were sacrificed and their kidneys were removed and prepared for light and electron microscopic examination.Kidneys in ciprofloxacin treated group by light microscope showed plenty of inflammatory cells in the interstitium (interstitial nephritis(with invasion of the tubules by inflammatory cells consist to with tubulitis.Also signs of degeneration of the kidney tubules were present in the form of cloudy swelling associated with focal tubular necrosis with intraluminal necrotic epithelial debris.By electron microscope the interstitial space between the proximal convoluted tubules were infiltrated by inflammatory cells and the basement membrane of the tubules become thickened.Also there was shortening of the folding of basal plasma membrane with few number of mitochondria that scattered in the cytoplasm which also contains mulliple vacuoles.There was loss of microvilli from the free surface of the cell.Acute interstitial nephritis due to quinolone therapy possibly may be undiagnosed in several cases bacause of the gradual onset of the symptoms,lack of oliguria the absence of any effect on the circulatory system.Clinicians should be aware of quinolone - associated acute interstitial nephsitis , which is a rare but potentially dangerous renal complication.
