Abstract:

Valsartan is a member of angiotensin type 1 receptor antagonists. The present study was carried out to screen the pharmacological effects of valsartan on isolated rabbit aortic strip and isolated rabbit heart. Also, the effect of valsartan on mean arterial blood pressure response to angiotensin(Ang) II injection was evaluated. Moreover, the cardio-protective effect of valsartan on acute myocardial infarction induced experimentally in rats was studied. Data obtained in the present study pointed out that valsartan produced significant decrease (P<0.05) in the Ang II induced contraction of isolated rabbit aortic strip but did not affect norepinephrine induced contraction in the isolated rabbit aortic strip. Also , valsartan in gradually increasing doses produced no effect on amplitude of contractility of isolated rabbit heart ,but significantly reduced the positive inotropic action of Ang II on isolated rabbit heart. Moreover, valsartan inhibited the pressor response to Ang II ir vivo as it lowers the elevated blood pressure induced by Ang II injection.Also, valsartan has cardioprotective effect on experimentally induced myocardial infarction induced by isoprenaline in rats. These results suggest a new potential for valsartan in the therapy of ischemic heart disease and chronic congestive heart failure but need further clinical investigations
