Abstract:

Ch lamydop kilo pnettmoniae (Cpneumoniae) is a common respiratory pathogen of human that has been linked to variety of chronic diseases at other body sites including atherosclerosis. It must have a mechanism by which it reaches these sites. The circulating monocytes are likely the vehicles for dissemination of C.pneumoniae from the site of primary infection to the site of the developing disease. Objective: The present work aimed at detection of viable infectious Cpneumoniae in the blood cells of normal blood donors (NBD), assessment of the normal donors for Cpneunzoniae carriage in circulating peripheral blood mononuclear cells (PBMNCs), and testing the ability of Cpneumoniae infected cell to transmit infection to host cell monolayer. Materials and Methods: 38 peripheral blood specimens from NBD were collected. Sera of the patients were tested for C.pneumoniae IgG by Enzyme Linked Immuno-Sorbent Assay (ELISA). PBMNCs fraction of the blood samples were immunofluorescent- stained for microscopic identification of Cot:eau:office-infected cells. Isolated white blood cells (WBC) were assayed by in vitro culture to evaluate infection transmission of blood cell borne Cpneuntoniae. Results: Using ELISA assay, 17/38 (44.7%) of NBD samples were positive IgG specific antibodies against C.pneumoniae. Imunofluorescence microscopy of smears demonstrated that 7/38(18.4%) of samples contained intracellular C pneuntoniae. In vitro culture demonstrated that WBCs of 9/38 (23.7%) of the NBD samples harbored infectious C pneuntoniae, that is clinically a potentially source of transmission. Conclusion: C. pneumoniae is present within peripheral blood cells of NBD, It was present in lysates of these peripheral WBC, and can be infectious and readily transmitted it to host cell monolayer.
