Forty eight adult male albino rats were used in this study. They
were divided into five groups; four of them (each consisted of ten rats)
were given the therapeutic dose of terbinafine and the fifth (consisting of
eight rats) was used as control. The first group was given the drug for
Iwo weeks, the second for six weeks and the third for twelve weeks. The
fourth group was given the drug for twelve weeks and left for another
month without drug to test the effect of drug withdrawal.
On light microscopic examination of liver specimens, vaculation of
the hepatocytes, dilatation of the blood vessels and infiltration of the
areas surrounding central veins and portal tracts with inflammatory cells
were noticed in the first three groups. These changes increase with the
duration of drug intake.
Ultrastructural examination showed some changes in the
mitochondria, endoplasmic reticulum, lysosomes and lipid droplets of the
hepatocytes in addition to mild dilatation of the bile canaliculi.
All light and electron microscopic changes disappeared after one
month of drug withdrawal. |
