Abstract:

The aim of this work is evaluation of the safety and efficacy of esmolol hydrochloride in attenuation of sympathetic response to endotracheal intubation as well as experimenttal evaluation of the haemodynamic effects of the drug in intact anesthetized rats for evaluation of the effect of gradually increasing doses of esmolol on heart rate and blood pressure, effect of esmolol on isoprenaline induced hypotension and tachycardia, effect of esmolol on adrenaline induced arrhythmia and drug interaction between esmolol and morphine. In all experiments heart rate and blood pressure were recorded. The results of these experiments revealed that esmolol had a significant dose-related reduction of blood pressure and heart rate when given by I.V. bolus injection, as well as esmolol was effective in controlling adrenaline induced arrhythmia, also there was interaction between morphine and esmolol where morphine potentiated effects of esmolol. In clinical work, the safety and efficacy of esmolol hydrochloride in attenuation of sympathetic response to endotracheal intubation was investigated on 100 patients of both sex, aged 20-40 years, divided into 5 equal groups. Group I: control group received 15m1 saline, Group II: received I.V. bolus dose of esmolol 1 mg/kg completed to 15 ml by saline. Group III: received I.V. bolus dose of esmolol 2 mg/kg completed to 15 ml by saline. Group IV: received I.V. bolus dose of fentanyl 2 pg/kg completed to 15 ml by saline. Group V: received I.V. bolus dose of esmolol 1.5 mg/kg plus fentanyl 2 jig/kg completed to 15 ml by saline
