CYP19 is a member of cytochrome P450 aromatase family, which regulates estrogen biosynthesis in
animals so it can be used as a candidate gene for marker-assisted selection strategies for fertility in
buffaloes. The sequence of this gene in buffalo was recently published but association study has not
conducted yet. Therefore, the objective of this study was to detect polymorphisms in a CYP19 locus
containing exon 7, and to determine associations between these polymorphisms and anestrum in
Egyptian buffalo. A PCR product of 241bp, containing exon 7 of CYP19 gene was successfully
amplified using PCR and subsequently, subjected to single strand conformation polymorphism (SSCP)
followed by sequence analysis to identify its different allelic patterns. There was no any SNP in all
examined Egyptian buffaloes as shown by one SSCP pattern and nucleotide sequencing. Consequently,
these results indicated that this locus does not influence anestrum in Egyptian buffalo and further studies
should be conducted to detect other SNPs associated with fertility in Egyptian buffalo. |
