Abstract:

Choline dehydrogenase (CHDH) of Ps. aeruginosa EG. isolate was showed to have enhanced hypersalinty. The genomic library of Ps .aeruginosa was isolated and screened for CHDH enzyme gene, The bet A gene code for CHDH enzyme cloned in PGEM-T easy vector and showed potential positive clone when expressed in E.coli DH5a.The size of gene insert with M.W. 1.7Kbp, When cut with restriction enzymes. Pilot protein expression study showed specific protein band of M.W. 60 KDa when expressed in E.coli DH5a, That induced with IPTG and isolated from parent strain, Finally it was demonstrated that the expression of the betA gene in Ps.aeruginosa was induced by the presence of salt, glucose, or choline precursor and IPTG inducer.
