Background:Thetobaccoepidemicsignifiesamajorpublichealththreat.Nicotine(NIC),amajoractiveconstituentintobacco,impedesmalefertilityandsemenquality.Thisworkisimplementedtoexplorethepotentialofseleniumnanoparticles(SeNPs)andthenewlyfabricatedSeNPs@vitaminC(SeNPs@VITC)nanocompositeinmitigatingtesticulartoxicityinducedbyNIC.Materialsandmethods:Thesixgroupsof48adultWistarratsweredesignedasfollows:thecontrolgroupinjectedintraperitoneallywithnormalsaline,theSeNPsgrouptreatedorallywith2mg/kgofSeNPs,theSeNPs@VITCnanocompositegrouptreatedorallywith2mg/kgofSeNPs@VITCnanocomposite,theNICgroupinjectedintraperitoneallywith1.25mL/kgofNIC,theNIC+SeNPsgroupreceivedSeNPsplusNIC,andtheNIC+SeNPs@VITCnanocompositegroupreceivedSeNPs@VITCnanocompositeplusNIC.Treatmentswereadministeredovera28-dayperiod.Results:NICtreatmentsignificantlycausedpoorspermquality,decreasedserumtestosterone,increasedfollicle-stimulatinghormone(FSH),luteinizinghormone(LH)concentrations,reducedhemoglobinlevels,leukocytosis,disruptedtesticularoxidant/antioxidantbalance,anddisorganizedtesticularstructure.TheconstructionofthenovelSeNPs@VITCnanocomposite,comparedtoNICplusSeNPsalone,demonstratedamorepotentameliorativeeffectonNIC-inducedreproductivetoxicityinadultrats.TheSeNPs@VITCnanocompositesignificantlyincreasedspermcount,reducedthepercentageofspermheadabnormalities,loweredbothserumFSHandLHconcentrations,andimprovedthehemoglobinresponse.Conclusions:BothSeNPsandSeNPs@VITCnanocompositealleviatedthetesticulartoxicityinducedbyNIC,buttheSeNPs@VITCnanocompositeexhibitedsuperiorefficacy.TheSeNPs@VITCnanocompositecouldbeemployedtoadvanceenhancedtherapeuticstrategiesforaddressingmaleinfertility. |
